چکیده

Cytotoxic T lymphocyte (CTL)-induced programmed cell deaths, including apoptosis and necroptosis, occur in skin diseases such as graft versus host disease (GVHD) severe drug eruptions. It has been reported that Fas-FasL, perforin, granzyme B induce these tumor necrosis factor-α (TNF-α) signaling is involved necroptosis. However, the molecular mechanisms of TNF-α deaths induced by CTLs have yet to be fully understood. Recently, a novel death pathway, PANoptosis, integrates all pyroptosis, apoptosis, necroptosis proposed, while its involvement GVHD still unknown. To address this gap knowledge, we administered inhibitor, etanercept, murine GVHD-like model with Ivl-mOVA mice generated. These transgenic express membrane-bound ovalbumin (mOVA) specifically epidermis under control involucrin (Ivl) promoter. By transfer OVA-specific CD8+T cells from OT-I mice, developed dermatitis day 7, half them died 21. The administration etanercept ameliorated improved survival rate. Flow cytometry annexin V staining detected many apoptotic keratinocytes on 14, were reduced treatment. Interestingly, an z-VAD, failed improve mortality, PANoptosis necrostatin-1 (nec-1), mortality. results indicate keratinocyte are via PANoptosis. interferon-γ trigger synergistically, mRNA levels cytokines increased ear mice. Our study demonstrates TNF-α-mediated controls during CTL-related dermatosis could therapeutic target for PANoptosis-related diseases.

TNF-α priming enhances CD4+FoxP3+ regulatory T-cell suppressive function in murine GVHD prevention and treatment.

CD4(+)CD25(+)FoxP3(+) regulatory T cells (Tregs) have been shown to effectively prevent graft-versus-host disease (GVHD) when adoptively transferred in murine models of hematopoietic cell transplantation and in phase 1/2 clinical trials. Critical limitations to Treg clinical application are the paucity of cells and limited knowledge of the mechanisms of in vivo function. We hypothesized that in...

Evasion of cytotoxic T lymphocytes by murine cytomegalovirus.

Murine cytomegalovirus causes lifelong infections with little pathology in normal host animals. Control of viral replication and prevention of pathology depend on both innate and adaptive immune mechanisms, and cytolytic T lymphocytes play a key role in this process. The virus encodes a number of genes which alter the normal assembly of class I major histocompatability complex proteins, and thu...

Cell Surface Glycoproteins of Murine Cytotoxic T Lymphocytes

The immune system is endowed with great flexibility. It has at its disposal a large number of alternative pathways of reaction against foreign, immunogenic substances. A sizeable part of this impressive flexibility is due to the fact that a number of subsets of specialized cell types coexist and interact within this system. Among these cell types, lymphocytes have been shown to play a dominatin...

Cytotoxic T lymphocytes

Cancer stem cells (CSCs)/cancer-initiating cells (CICs) are characterized as a small population of cancer cells that have high tumor-initiating ability. CSCs/CICs are resistant to several cancer therapies, and eradication of CSCs/CICs is essential to cure cancer. How can we eradicate CSCs/CICs? Cytotoxic T lymphocytes (CTLs) might be a promising answer.

Resistance of cytotoxic T lymphocytes to lysis by a clone of cytotoxic T lymphocytes.

To investigate how cytotoxic T lymphocytes (CTL) avoid killing themselves when they destroy target cells, we compared 20 different cell lines as target cells, including several CTL cell lines, for their susceptibility to lysis by CTL. Variations in recognition of this diverse set of target cells was circumvented by attaching to all of them a monoclonal antibody to the antigen-specific receptor ...
